Aneuploidy and its relationship with growth in different populations of the Pacific oyster (Crassostrea gigas, Thunberg) ArchiMer

Cytogenetic abnormalities arising both in mitosis and meiosis are known to be common in bivalves. Here we review result obtained from the observations of somatic aneuploidy in different populations of the commercially important Pacific oyster, Crassostrea gigas. The oysters studied were either (1) produced in IFREMER hatcheries between 1986 and 1999, (some of these were part of the EU funded programme GENEPHYS); or (2) collected from wild populations along the French Atlantic coast. Autotetraploid Pacific oysters (Crassostrea gigas) obtained using normal diploid eggs: induction and impact on cytogenetic stability ArchiMer

We describe two methods of producing viable and fertile autotetraploid Pacific oyster (Crassostrea gigas Thunberg) based on the use of normal-sized oocytes produced by normal diploid females. Our methods showed that the oocyte size is not a limiting factor for the success of the induction to autotetraploidy. These methods offer means of direct introgression of genetic progress from elite diploid lines to tetraploids used as broodstock, avoiding a triploid step with the risk of transferring undesirable traits from highly fecund triploids. Pacific oysters, Crassostrea gigas, are commonly reared in estuaries where they are exposed to anthropogenic pollution. Much research has been made on the toxicity of cadmium to aquatic organisms because the compound recurrently contaminates their environment. Our study examined the influence of cadmium on aneuploidy level (lowered chromosome number in a percentage of somatic cells) and hemocyte parameters in C. gigas at different stages of life. Evidence of differential chromosome loss in aneuploid karyotypes of the Pacific oyster, Crassostrea gigas ArchiMer

The G-banding technique was performed on aneuploid karyotypes from gill tissue of the Pacific oyster, Crassostrea gigas, to assess whether chromosome losses could be explained by differential chromosomal susceptibility and to clarify the negative correlation between aneuploidy and growth rate previously reported in different populations of this oyster. The study of 95 G-banded aneuploid karyotypes showed that only 4 of the 10 chromosome pairs (viz. 1, 5, 9, and 10) of C. gigas were affected by the loss of one homologous chromosome. Pairs 1, 9, and 10, which were lost in 56, 33, and 44% of cases, respectively, may be considered to be differentially affected. Hypotheses on this differential chromosomal susceptibility are discussed.

Experimental evidence for a genetic basis to differences in aneuploidy in the Pacific oyster (Crassostrea gigas) ArchiMer

Aneuploidy has been previously reported in wild and cultivated Pacific oysters, Crassostrea gigas, and has been shown to be negatively correlated with growth. This is especially important since high variability of growth rate is one of the major problems in the aquacultural production of this species. The existence of a genetic basis for the observed differences in aneuploidy was first investigated through the comparative study of six full-sib families, with mean individual weights ranging from 0.59 to 1.49 g. The slowest growing family was found to have the highest level of aneuploidy. In diploid Pacific oysters, aneuploidy affects 1 to 3 chromosomes in up to 35% of the somatic cells and is positively correlated with inferior growth of the animal and thus could be associated with significant economic loss in this major aquaculture species. Genetic transmission of Solanum demissum (2n=6x=72) chromosomes from a pentaploid hybrid of S. tuberosum (2n=4x=48) into the aneuploid BC1 progeny OAK

Solanum demissum is a homozygous, hexaploid wild species (2n = 6x = 72, AADDD(d)D(d)) that has long been used as a source of late blight resistance by repeated backcrossing with S. tuberosum (2n = 4x = 48, AAA(t)A(t)). To understand how S. demissum germplasm is incorporated to S. tuberosum through backcrosses, a linkage analysis of 590 DNA markers from S. demissum-specific simplex alleles was performed in an aneuploid population of 87 BC1 plants. Genetic variation among and within tritipyrum (Thinopyrum bessarabicum x Triticum durum) lines using PCR-based molecular markers Ciencia e Investigación Agraria

Primary hexaploid tritipyrum lines are amphiploids between Triticum durum and Thinopyrum bessarabicum, which can set seed in at least 250 mM NaCl. Genetic variation within and among 14 lines of primary tritipyrum were investigated by using 2 random and 6 semi random primers that produced clear and reproducible DNA amplicons. Average values for Nei's gene diversity (h) and Shannon indices (I) were 0.115 and 0.178, respectively. The impact of environmental factors on aneuploidy level in the oyster Crassostrea gigas was studied with experiments in controlled conditions which exposed adult and juvenile oysters to different pollutants (atrazine, a herbicide, and cadmium, a heavy metal) frequently found in Marennes-Oléron Bay. A positive correlation between atrazine concentration and aneuploidy in C. gigas was observed. Moreover, the impact of atrazine on aneuploidy level persisted in time both within and between generations, demonstrating the influence of an environmental pollution on this phenomenon. In the Pacific oyster, Crassostrea gigas hypodiploid aneuploid cells with 2n=19, 18 or 17 chromosomes have regularly been reported. There is a negative correlation between somatic aneuploidy and growth rate in oysters (Leitão et al., 2001a) as well as evidence for a genetic basis (Leitão et al., 2001b). Impact of diuron on aneuploidy and hemocyte parameters in Pacific oyster, Crassostrea gigas ArchiMer

Diuron is a substituted urea herbicide used for agricultural and nonagricultural weed control. Its widespread use and relatively slow breakdown led us to analyze its influence on aneuploidy level (lowered chromosome number in a percentage of somatic cells) and hemocyte parameters in Pacific oysters, Crassostrea gigas. Adult oysters were subjected to two diuron concentrations (300 ng L-1 and 3 mu g L-1) for 11 weeks. Vincristine induces somatic segregation, via mitotic crossing-over, in diploid cells of Aspergillus nidulans Biol. Res.

Vincristine is an alkaloid widely used as an antineoplastic agent. In eukaryotic cells the drug causes blockage in the G2 phase of the cell cycle and an increase in the frequency of sister chromatid exchanges. Due to the fact that germinating Aspergillus nidulans cells spend most of their cycle in G2 phase, they provide an excellent system for the study of mitotic crossing-over. Taking into account that mitotic crossing-over occurs during G2 period, the evaluation of recombinagenic and aneugenic potential of vincristine is provided with regard to two diploid strains of A. nidulans: a wild strain (uvsH+//uvsH+) and a defective one in DNA repair (uvsH//uvsH).
